Ego Pharmaceuticals' Braeside site will see a 50 per cent increase in production capacity by 2027.
It will also create 90 new jobs for the Victorian site, which already employs about 400 people.
The state government is supporting the project but its funding commitment is under commercial confidence.
"Ego Pharmaceuticals' decision to expand its operations in Victoria demonstrates the strength of our nation-leading advanced manufacturing industry and highly skilled workers," Industry Minister Ben Carroll said in a statement.
"We'll continue to back local manufacturers because it builds our capacity to make more things in Victoria and creates secure jobs."
Ego Pharmaceuticals produces 150 dermatological skincare products for Australian and international markets, with its brands including QV, SunSense, Elucent and Aquim.
It started operating 70 years ago out of an Elsternwick home and has since grown into a global brand with more than 730 employees in 15 countries.
"Keeping our high-technology manufacturing in Australia has and always will be important to us," Ego Pharmaceuticals managing director Alan Oppenheim said in a statement.
"This (government) grant will help us further in creating good jobs to support our local economy as we continue to grow and will enable Ego to transform more lives through the science of healthy skin across many nations."
